Your title is "ESP to ESP handshaking" so you must have something connecting the two devices! Either wire or wireless. I guess you could use a laser beam and a light detector to give simplex one-way but for duplex you could wire Rx to Tx and Tx to Rx to get bidirectional comms.
Therefore if you want to use the 2.4G wireless capability instead of stringing wire, you need a wireless protocol to connect the two devices.

I would just click controlled relays. Like "Blink", including feedback control.
